چکیده

background: high-grade astrocytoma (grade 4) or glioblastoma multiforme (gbm) are deadly brain tumors. new therapies attempt to increase lifetime and quality of life in patients with malignant astrocytoma. o6-methylguanine-dna methyltransferase (mgmt) enzyme expression may be effective in prognosis and response to treatment of these patients. the aim of this study was assessment of mgmt enzyme expression in patients with astrocytoma grade 4. materials and methods: in this study, 48 patients with gbm that were treated with surgery, chemotherapy and radiotherapy were investigated and followed-up for 47 months for the survival rate. pathology blocks of patients were examined for mgmt enzyme expression using immunohistochemistry method. results: the patients were 34 males and 14 females. the ages ranged from 24 to 77 years, with a mean age of 53.52 â± 13.39 years. there was no significant difference between two groups (positive and negative mgmt enzyme expression) in overall survival (median [range]11.5 [4-30] vs. 13 [5-22], p = 0.9). the results of our study showed that patients although who were undergone near total surgery had higher overall survival than the group of patients who had biopsy only however, it was not significant. patients who were treated with temozolomide (tmz) (temodal, merck canada) had significant overall median survival (14.5) more than the patients who were treated with procarbazine (roche, swiss)-lomustine (lilly, usa)-vincristine (lilly, usa) regimen (8.75) (p < 0.05). conclusion: o6-methylguanine-dna methyltransferase enzyme expression had no effect on survival of patients with grade 4 brain astrocytoma tmz may increase survival rate.

Evaluation of O6-methylguanine-DNA methyltransferase enzyme expression effect on survival of patients with Grade 4 brain astrocytoma

BACKGROUND High-grade astrocytoma (Grade 4) or glioblastoma multiforme (GBM) are deadly brain tumors. New therapies attempt to increase lifetime and quality of life in patients with malignant astrocytoma. O6-methylguanine-DNA methyltransferase (MGMT) enzyme expression may be effective in prognosis and response to treatment of these patients. The aim of this study was assessment of MGMT enzyme e...

Prognostic significance of loss of O6-methylguanine-DNA methyltransferase expression in supratentorial diffuse low-grade astrocytoma.

BACKGROUND O(6)-Methylguanine-DNA methyltransferase is a DNA repair protein. Epigenetic silencing of MGMT function by its promoter hypermethylation is considered to contribute to carcinogenesis. If loss of function in MGMT is related to tumor progression, the immunohistochemical method may predict the malignant change of gliomas. METHOD We investigated the expression of MGMT by immunohistoche...

Ribozyme-mediated modulation of human O6-methylguanine-DNA methyltransferase expression.

A synthetic oligonucleotide containing ribozyme sequences targeted to the 5' region of the human O6-methylguanine-DNA methyltransferase (MGMT) mRNA has been constructed. This ribozyme demonstrates cleavage activity in vitro in the presence of Mg2+. To determine whether this ribozyme can function in vivo, HeLa CCL2 cells were transfected with a mammalian expression vector containing the ribozyme...

DNA repair enzyme, O6-methylguanine DNA methyltransferase, modulates cytotoxicity of camptothecin-derived topoisomerase I inhibitors.

Two camptothecin-resistant cell lines, CPT30 and KB100, were established and characterized previously in our laboratory. Because enhanced sensitivity to 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) and decreased expression of O(6)-methylguanine-DNA methyltransferase (MGMT) protein were observed in these lines, we hypothesized that MGMT may be a determinant of cytotoxicity associated with camptot...

Inhibition of human O6-methylguanine-DNA methyltransferase by 5-methylcytosine.

The ability of cloned human O6-methylguanine-DNA methyltransferase to repair a methylated guanine in a CpG-containing sequence, i.e., island, was studied by using a synthetic double-stranded 20-mer oligonucleotide from codon 248 of the p53 gene and another designed sequence. The double-stranded oligonucleotides incorporating 5-methylcytosine (5mC) and O6-methylguanine (O6mG) in various combinat...
